Провести валидацию и оценить точность 4 шкал генетического риска (ШГР) артериальной гипертензии (АГ), созданных ранее на европейских выборках, на популяционной выборке населения Ивановской области.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Для генетического анализа было использовано секвенирование следующего поколения с использованием таргетной панели на выборке региона Центральной России (n=1682) на основе коллекции биобанка. Для валидации отобрано четыре ШГР, ассоциированных с АГ, ранее разработанных для европейской популяции. В качестве метрик качества для моделей регрессии использованы коэффициент детерминации и площадь под ROC-кривой. Проведена дополнительная валидация с включением всех вариантов нуклеотидной последовательности, независимо от уровня их неравновесного сцепления. Составлена объединенная ШГР на основе коэффициентов из отдельных ШГР методом C+T (clumping + thresholding).</p></sec><sec><title>Результаты</title><p>Результаты. В рамках исследования было продемонстрировано, что предсказательная способность ранее разработанных ШГР при использовании для популяции региона Центральной России ниже, чем в оригинальных исследованиях, доля объясненной дисперсии составила 0,5-0,8%. Наилучшую предсказательную способность (доля объясненной дисперсии — 2,5%) продемонстрировало применение ранее разработанной ШГР (Evangelou E, et al., 2018), включающей наибольшее количество вариантов нуклеотидной последовательности (n=852).</p></sec><sec><title>Заключение</title><p>Заключение. ШГР АГ, разработанные на европейских выборках, не рекомендуется использовать у представителей российской популяции без предварительной валидации. Для создания собственных ШГР не рекомендуется объединять статистические параметры (β -коэффициенты и p-value) из различных ШГР с целю создания собственных ШГР.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. To validate and evaluate the accuracy of 4 genetic risk scores (GRSs) for hypertension (HTN), previously created on European samples, on a population sample of the Ivanovo Oblast.</p></sec><sec><title>Material and methods</title><p>Material and methods. For genetic analysis, targeted next-generation sequencing was used on a sample of the Central Russia (n=1682) based on the biobank collection. Four GRSs associated with HTN, previously developed for the European population, were selected for validation. The coefficient of determination and the area under the ROC curve were used as quality metrics for regression models. Additional validation was carried out to include all nucleotide sequence variants, regardless of linkage disequilibrium level. A combined GRS was compiled based on coefficients from individual GRSs using the clumping + thresholding (C+T) method.</p></sec><sec><title>Results</title><p>Results. The study demonstrated that the predictive value of previously developed GRSs when used for Central Russian population is lower than in the original studies. The proportion of explained variance was 0,5-0,8%. The best predictive ability (proportion of explained variance — 2,5%) was demonstrated using previously developed GRSs (Evangelou E, et al., 2018), which includes the largest number of nucleotide sequence variants (n=852).</p></sec><sec><title>Conclusion</title><p>Conclusion. GRSs for HTN, developed on European samples, is not recommended for Russian population without preliminary validation.
