Оценить допустимость факта прохождения неинвазивного пренатального тестирования (НИПТ) в роли информативного критерия для определения качества плазмы крови (ПК) и внеклеточной дезоксирибонуклеиновой кислоты (ДНК) (вкДНК) в случае, когда на преаналитическом этапе используются пробирки со стабилизирующим составом, на примере образцов ПК, подвергнутых продолжительному хранению при комнатной температуре (+18о С) и многоцикличному замораживанию-оттаиванию.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Образцы ПК подвергали 20 циклам замораживания-оттаивания (-80о С/+18о С), 20-дневному хранению при температуре +18о С с промежуточной оценкой состояния вкДНК. Количественный выход оценивали флуорометрически, степень фрагментации — с помощью полимеразной цепной реакции (ПЦР) в реальном времени, результаты НИПТ — через высокопроизводительное секвенирование.</p></sec><sec><title>Результаты</title><p>Результаты. После многоцикличного замораживания-оттаивания и длительного хранения ПК при комнатной температуре (+18о С) наблюдается снижение целостности вкДНК, уменьшение концентрации вкДНК к 20 циклу оттаивания и тенденция к увеличению концентрации к 20 сут. хранения ПК; несмотря на это, результаты НИПТ исследованных образцов показали высокую степень совпадения с результатами НИПТ референсных образцов.</p></sec><sec><title>Заключение</title><p>Заключение. Показано, что один только факт успешного проведения НИПТ не может рассматриваться в качестве надежного и достаточного критерия оценки качества исходной ПК и правильной преаналитики. Это подчеркивает особую важность контроля условий транспортировки и хранения образцов ПК и цельной крови.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. To assess non-invasive prenatal testing (NIPT) as an informative criterion for quality of blood plasma and cell-free deoxyribonucleic acid (DNA) (cfDNA) in the case of using stabilization tubes at the preanalytical phase, in the example of plasma samples subjected to long-term storage at room temperature (+18о С) and multiple freezethaw cycles.</p></sec><sec><title>Material and methods</title><p>Material and methods. The plasma samples were subjected to 20 freeze-thaw cycles (-80о С/+18о С), 20-day storage at +18о С with an intermediate cfDNA assessment. The quantitative yield was assessed by fluorometry, while the fragmentation and NIPT data — using realtime polymerase chain reaction (PCR) and high-throughput sequencing, respectively.</p></sec><sec><title>Results</title><p>Results. After multiple freeze-thaw cycles and long-term plasma storage at room temperature (+18о С), a decrease in the integrity and the concentration of cfDNA by the 20th thawing cycle, as well as a tendency to an increase in concentration by 20 days of storage were observed. Despite this, the NIPT results of the studied samples showed a high degree of coincidence with the NIPT data of the reference samples.</p></sec><sec><title>Conclusion</title><p>Conclusion. The mere fact of successful NIPT cannot be considered as a reliable and sufficient criterion for assessing the quality of initial plasma and correct preanalytics.
